AN EXPERIMENTAL STUDY OF FIBROSARCOMA ORIGINATED FROM PERIODONATL LIGAMENT


Abstract
-Abstract-
The purpose of this study was to investigate induction of odontogenic fibrosarcoma by
injection of DMBA into pulp canal.
Fifty male hamsters used in this experiment, weighing 100-200gm were devided into
two groups. In experimental group, forty animals were cut right mandibular anterior
tooth at the gingival margin level with surgical bur. After pulp tissue were removed
and pulp canal were enlarged with endodontic files, 0.1-0.2ml of 0.5£¥ DMBA solution
which was dissolved in mineral oil was injected into pulp canal. Subsequently, pulp
canals were temporarily filled with zincoxide eugenol cement. In control group, ten
animals were injected with mineral oil by the same method.
Two animals in control group and fie animals in experimental group were sacrificed at
6, 10, 12, 16, 20 weeks after experiment, respectively. When sacrificed, the right
mandible was removed and fixed in 10£¥ N-formalin. After decalcification with 5£¥ nitric
acid solution, they were embedded with paraffin and sectioned by routine method. They
were stained with H-E, PAS, Van Gieson, and Masson's trichrome.
The results were as follows ;
1. Most of tooth germinal tissue were necrosis forming abscess, gradually changed to
granulation tissue, and then replaced by connective tissue. Ameloblasts in apical portion
were proliferated into irregular stratified form.
2. At sis and ten weeks, periodontal ligament of apical portion showed granulation tissue
or proliferation of connective tissue, and initiated proliferation of fibrosarcoma. Tumor
cells infiltrated and proliferated into alveolar bone, and into peripheral muscular and
glandular tissue.
3. At 12 weeks, 2 cases showed ameloblastomatous epithelial proliferation in apical
portion.
4. 2 cases showed squamous cell carcinoma combined with fibrosarcoma.
5. AT PAS stain, periodontal ligament was poorly positive, and residual alveolar bone
was positive, but fibrosarcoma was negative. In proliferative fibrosarcoma, collagen
fibers did not showed the positive staining of Van Gieson and Masson trichcome.
